2. Synthesis of 4-amino-4,6-dideoxyglucose: D-glucose-1-phosphate is converted into dTDP-4-amino-4,6-dideoxy-D-glucose through nucleotidylation, dehydration, and transamination.


3. Synthesis of acarviosyl moiety: NDP-1-epi-valienol-7-phosphate and dTDP-4-amino-4,6-dideoxy-D-glucose undergo sugar transfer reaction mediated by glycosyltransferase to form dTDP-acarvicose-7-phosphate, which then directly combines with maltose to generate acarbose-7-phosphate. Acarbose-7-phosphate is synthesized inside the cell, but to finally produce acarbose, it needs to be transported outside the cell via the transmembrane protein AcbWXY/GacWXY and undergo dephosphorylation.
